眼针对急性脑缺血再灌注损伤大鼠海马组织BDNF表达影响

摘　　要：目的:观察眼针对急性脑缺血再灌注模型大鼠海马脑源性神经因子(BDNF)表达的影响,探讨眼针对急性脑缺血再灌注损伤治疗的机制。方法:应用线栓法复制急性脑缺血再灌注损伤大鼠模型,采用眼针治疗,于再灌注后3 h进行神经功能缺损评分,采用Western blot方法、实时荧光定量聚合酶链反应(RQ-PCR)方法测定海马BDNF蛋白及mRNA表达的变化。结果:与模型组比较,眼针组大鼠神经功能缺损评分明显下降,眼针组大鼠海马组织BDNF蛋白及mRNA表达明显上调(P<0.05)。结论:眼针具有明显的改善大鼠脑缺血再灌注损伤的作用,其机制与上调海马组织BDNF表达有关。Objective：To investigate the effect of the eye acupuncture therapy on hippocampus BDNF expression of rats with acute cerebral ischemia-reperfusion injury and the related mechanism. Methods：To establish the rat model of cerebral ischemia-reperfusion by suture method.After reperfusion 3h,the neurophysical behaviours were accessed by ZeaLonga neurophysical impairment marks.The methods of Western-blotting and RQ-PCR were taken to detect the alteration of the BDNF protein and BDNF mRNA in rat hippocampus after the eye acupuncture therapy. Results：Compared with the model group,the neurologic impairment score of the eye-acupuncture group decreased obviously;The expression of BDNF protein and BDNF mRNA in rats hippocampus after the eye acupuncture therapy was strongly increased（P0.05）. Conclusion：The eye acupuncture therapy can improve the cerebral ischemia-reperfusion injury evidently by increasing the expression of the hippocampus BDNF protein.
