竹节参总皂苷对环磷酰胺致免疫低下小鼠免疫功能的影响

摘　　要：目的研究竹节参总皂苷(Total Saponins of Panax japonicus Rhizoma,TSPJ)对环磷酰胺诱导的免疫低下小鼠免疫功能的影响。方法腹腔注射环磷酰胺(Cy)复制免疫功能低下的动物模型;摘取脾脏称质量并计算脾脏指数,MTT法检测T淋巴细胞增殖活性,微量分光光度法检测血清溶血素抗体HC50形成水平,流式细胞术检测外周血T淋巴细胞亚群(CD4+T细胞、CD8+T细胞)比例、B细胞及NK细胞比例,ELISA法检测血清IFN-γ量。结果 TSPJ可增加免疫低下小鼠脾脏质量与脾脏指数,促进脾脏T淋巴细胞增殖,提高免疫低下小鼠血清溶血素水平,上调外周血CD4+/CD8+比例、B淋巴细胞及NK细胞比例,提高血清中IFN-γ的水平。结论 TSPJ通过刺激小鼠的特异性及非特异性免疫应答,对环磷酰胺致免疫低下小鼠的免疫功能有良好的增强作用。AIM To investigate the immunomodulatory effects of total saponins of Panax japonicus Rhizoma（TSPJ） on cyclophosphamide（Cy）-induced immunosuppressed mice.METHODS In this study,immunosuppressed murine models were established in Kunming mice induced by Cy,the splenic indexes were evaluated,the percentage of CD4＋,CD8＋ T cells,B cells,NK cells and ratios of CD4＋/CD8＋ in peripheral blood were analysed by flow cytometry（FCM）.IFN-γ serum levels were detected by ELISA.Activity of splenic lymphocyte proliferation was assayed by MTT and levels of serum hemolysin were confirmed by spectrophotometry.RESULTS TSPJ evidently increased splenic index,improved the percentage of peripheral B cells,NK cells and ratios of CD4＋/CD8＋,raised serum levels of IFN-γ,and promoted the proliferation of splenic lymphocytes and the production of serum hemolysin.CONCLUSION TSPJ shows significant immunomodulatory property on immunosuppressive mice through specific and nonspecific immunity.
