反相高效液相色谱法测定白芷提取分离物中欧前胡素含量

摘　　要：目的建立测定白芷乙醇提取分离物中欧前胡素含量的高效液相色谱法。方法色谱柱为Zorbax Eclipse XDB-C18柱,流动相为0～6 min甲醇-水(65∶35),6～15 min甲醇-水(55∶45),15～25 min甲醇-水(65∶35),流速1 mL/min,检测波长302 nm,柱温为室温。结果欧前胡素质量浓度在66～330μg/mL范围内与峰面积线性关系良好,线性方程为Y=0.0142X+0.006,r=0.9998(n=5),平均回收率为99.50%,RSD=1.34%(n=15),3批白芷提取物中欧前胡素的含量分别为1.01,0.98,0.92 g/L。结论所用方法快速、简便,结果可靠,可作为大孔吸附树脂分离白芷乙醇提取物中欧前胡素的含量测定方法。Objective To establish a method for the determination of imperatorin in Angelica dahurica ethanol-extracts,isolated by macroporous absorbent resin.Methods RP-HPLC method was used.The column was Zorbax Eclipse XDB-C18 column,with methanol-water as a mobile phase using a gradient elution,the flow rate was 1.0 mL/min,imperatorin was determined at 302 nm,and the column temperatur e was room temperature.Results In the range of 66-330 μg/mL,imperatorin was l inear（r=0.999 8）,with an average recovery of 99.50%,RSD=1.34%（n=15）.Imperato rin content of 3 batches were 1.01,0.98,0.92 g/L,respectively.Conclusion The method is simple,fast and accurate for the analysis of imperatorin.
