提取细菌胞内PHB方法的研究

摘　　要：以芽胞杆菌作为实验材料,研究了SDS法、NaClO法、SDS-NaClO法、CHCl3法、CHCl3-CH3OH法等对细菌胞内PHB的提取率及纯度影响。结果表明,NaClO法为最佳的PHB提取方法,其提取率达57.3%,提取出PHB的性质和纯度与PHB标准样品基本一致。NaClO法的最佳提取条件为:提取温度45℃,NaClO浓度为30%,提取时间为20 min。The effect of SDS,NaClO,SDS-NaClO,CHCl3 and CHCl3-CH3OH method on extraction and purity of PHB were studied with Bacillus HM123761 as material.The results showed the suitable method was sodium hypochlorite,the rate of PHB extraction was 57.3%.The nature and purity of PHB was similar to the sample.The largerest quantity of PHB was obtained when the cells were treated by 30%（w/v） sodium hypochlorite for 20 min at 45 ℃.
