摘 要:目的:从尿激酶型纤溶酶原激活物(uPA)系统研究复元胶囊含药血清及其主要成分淫羊藿苷、三七皂苷R1治疗骨关节炎的分子机制。方法:原代培养兔软骨细胞,TNF-α诱导软骨细胞损伤,分为7组:空白组、模型组(TNF-α10μg.L-1组)、盐酸氨基葡萄糖25 g.L-1组、20%复元胶囊含药血清组、淫羊藿苷12.5 mg.L-1组、三七皂苷R1 125 mg.L-1组、淫羊三七组(12.5,125 mg.L-1),RT-PCR检测各组uPA,NF-κB(P65)mRNA的含量,EMSA检测NF-κB(P65)与DNA结合的活性,Western检测IκBα水平。结果:复元胶囊含药血清及其主要成分淫羊藿苷、三七皂苷R1均可显著降低uPA,NF-κB mRNA表达量,降低NF-κB(P65)与DNA结合的活性,升高IκBα水平,与TNF-α组比较,统计学有显著差异(P<0.01),各给药组间无明显差异。结论:复元胶囊含药血清及其主要成分淫羊藿苷、三七皂苷R1可通过降低NF-κB(P65)的活性,升高IκBα水平,从而降低uPA的表达,保护软骨细胞免受炎症因子的损伤。Objective: To study the molecular mechanisms of Fuyuan capsule serum containing, icariin and arasaponin R1 in the treatment of osteoarthritis from the urokinase-type plasminogen activator (uPA) system. Method: Chondrocytes were isolated, cul- tured and identified using type I1 collagens immunostaining. After stimulating with TNF-α 10 μg · L^-1, 1 h, then the ehondrocytes were treatment with glucosamine hydrochloride 25 g · L-l , 20% Fuyuan capsule serum c0ntaining, ieariin 12. 5 mg · L^-1 , arasaponin Rl 125 mg·L^-1 , icariin 12. 5 mg · L^-1 + arasaponin R1125 mg · L^-1. After 2 h, expression of uPA and nuclear factor kappa B (NF-κB P65 ) mRNA was detected by reverse transcription polymerase chain reaction (RT-PCR), the activities of NF-κB (P65) combine DNA were determined by electrophoretie mobility shift assays (EMSA), IκBα were detected by Western blotting. Result: Fuyuan capsule, ieariin and arasaponin R1 could significantly reduce NF-κB( P65 ) activities and uPA mRNA expression, and increase expression of IκBα(P 〈0. 01 ), but no significant difference between all treatment groups. Condusion: Fuyuan capsule and its two main active ingredients, ieariin and arasaponin R1, could protect chondrocytes from damage through reducing the NFκKB(P65) activities, in- creasing the express of IκBα and then reducing uPA of chondrocytes.
