HPLC测定紫草油中左旋紫草素的含量

摘　　要：目的建立紫草油中左旋紫草素的含量测定方法。方法采用HPLC法，色谱柱为Acclaim 120 C18（4．6×250mm,5μm）．流动相为甲醇．水．磷酸（78：22：0．3，v／v），检测波长为516nm，柱温为35℃。结果在8．56-42．80ug·mL^-1之间，左旋紫草素进样量和峰面积具有良好的线性关系．线性方程为y=o．4088x＋0．033。r=0．9999；测得紫草油中左旋紫草素平均含量为15．67pg·mL^-1。结论该方法简便可行，重现性好。可作为紫草油的质量控制方法。OBJECTIVE To establish the determination method of L-Shikonin in Shikonin oil. METHODS The column was Acclaim 120 C18（4.6×250mm, 5μm）, mobile phase was methanol-water-phosphoric acid（78：22 ：0.3, v/v）, the detection wavelength was 516nm, column temperature was 35℃. RESULTS L-shikonin sample volume and the peak area had a good linear relationship in 8.56 - 42.80μg·mL^-1, linear equation was y = 0. 4088x ＋ 0. 033, r = 0. 9999; the average content of L-Shikonin in Zicao oil was 15.67μg·mL^-1. CONCLUSION The method is simple, feasible, and reproducible and can be used as the quality control method of Zicao oil.
