摘 要:通过在毛细管聚合物整体柱表面修饰金纳米粒子,制备了一种可选择性捕获含巯基化合物的固相微萃取整体柱。首先制备聚(甲基丙烯酸缩水甘油酯-乙二醇二甲基丙烯酸酯)整体柱,在其表面化学修饰半胱胺;通过半胱胺上的巯基基团将金纳米粒子固定在整体柱的孔表面。以巯基类化合物为探针,评价了固相微萃取整体柱的萃取性能。结果表明,由于Au对巯基配体间有高亲和性,此柱能选择性捕获巯基化合物。金纳米粒子整体柱及其再生柱对谷胱甘肽的萃取容量分别为0.718和0.603μmol。考察了此微萃取整体柱对巯基药物的萃取富集性能,其对卡托普利的富集倍数达30倍,可用于血浆样品中卡托普利的萃取富集。A novel polymer monolithic capillary column functionalized with gold nanoparticles(Au NPs) that enabled the selective capture of thiol-containing compounds was prepared for solid phase microextraction.The column was prepared from a poly(glycidyl methacrylate-co-ethylenedimethacrylate) monolith and has been modified chemically with cysteamine,followed by immobilization of that Au NPs on the surface of the pores of the monolith.Thiol-containing compounds as model compounds were tested to evaluate the performance of this solid-phase microextraction monolith.The results indicated that the polymer monolith could selectively capture thiol-containing compounds from analytes because AuNPs have strong affinity to thiol-containing ligand.The loading capacity of the polymer monolith modified with Au NPs and the regenerated column determined for glutathione was 0.718 and 0.603 μmol,respectively.The extraction and enrichment of thiol-containing drug on polymer monolith modified with AuNPs was also investigated.It was found that the enrichment multiple was 30 folds for captopril and this monolith could be used for the extraction and enrichment of trace captopril in plasma.
