摘 要:目的利用小鼠胚胎干细胞检测生物珊瑚人工骨材料的细胞生物相容性。方法小鼠胚胎干细胞与生物珊瑚骨支架材料混合体外培养为实验组,设单纯小鼠胚胎干细胞培养为对照组,分别于第2、4、6、8天进行MTT法检测细胞增殖活性,特异性胚胎抗原-1检测及电镜扫描观察。采用SPSS13.0对所得数据行t检验。结果 MTT法检测细胞增殖活性结果显示,实验组在培养2d、4d时,与对照组组间无显著性差异(P>0.05),在6d及8d时实验组MTT值明显高于对照组,且组间具有显著性差异(P<0.05)。特异性胚胎抗原-1检测及电镜扫描证实小鼠胚胎干细胞对生物珊瑚人工骨支架材料具有良好的粘附性,在其三维微孔隙内能较快增殖。结论体外小鼠胚胎干细胞检测试验证实珊瑚人工骨具有良好的细胞生物相容性,具备骨组织工程支架材料的良好特性。Objective To test the cytocompatibility of biological coral artificial bone (BCAB) using mouse embryonic stem cells (MESCs) in vitro. Methods MESCs cultured with BCAB scaffold were given as experimental group and axenic cultivation of MESCs served as control group. The cell proliferation was detected by MTT method on the 2nd day, 4th day, 6thday and 8th day respectively. Stage specific embryonic antigen-1 (SSEA-1) detection and SEM observation was also performed at the same time. The data were compared and statistically analyzed with SPSS 13.0. Results The differences of MTT detection results between the two groups were not significant on the 2nd day and 4th day (P 0.05). But on the 6th day and 8th day, the MTT detection results of the experimental group were higher than those of the control group and there was significant difference between the two groups (P 0.05). The results of SSEA-1 detection and SEM observation demonstrated that MESCs had good bioadhension to BCAB scaffold and remained good proliferation in 3D-micropore space of BCAB scaffold. Conclusions The BCAB has good cytocompatibility with MESCs, which can be used as a kind of matrix material for bone tissue engineering.
