摘 要:目的观察不同途径移植骨髓间充质干细胞(BMSCs)对脊髓损伤修复效果。方法选取20g Wis-ter大鼠采用贴壁法分离、培养、纯化BMSCs,选取180gWister大鼠制备脊髓损伤模型动物,分别采用损伤部位和腰骶鞘内两条途径移植细胞,通过BBB评分和细胞形态学方法,观察不同途径移植BMSCs治疗脊髓损伤的效果。结果腰骶鞘内细胞移植后第2、3周进行BBB评分各时间段均较损伤部位细胞移植组评分高。另外形态学观察显示BMSCs经腰骶鞘内移植后组织结构规则,形成明显的网状结构,且组织内空泡数量明显少于BMSCs经损伤部位移植组。结论经腰骶鞘内移植BMSCs的疗效好于经损伤部位细胞移植。Objective To observe the different ways bone marrow mesenchymal stem cells(BMSCs) transplant on the effect of spinal cord injury repair.Methods BMSCs were isolated and purified from 20g of Wister rats by by adherent culture.180g of Wister rats were selected for preparation of model of spinal cord injury.BMSCs transplant adopted two pathways of injury parts or the lumbosacral scabbard transplant.The nerve conduction function,sports function and morphological examination were performed in different time after BMSCs transplant.Results After 3 weeks transplanted,BBB scores is higher in the lumbosacral scabbard transplant group than the site of injury transplant.The morphological observations indicate that the organizational structure appear clear network structure,and significantly less than the number of vacuoles within the organization in lumbosacral scabbard transplant group.Conclusion lumbosacral scabbard transplantation of BMSCs to repair SCI is more effective than through the injury site.
