摘  要:研究通过PCR-SSCP技术对大白猪、长白猪和杜洛克猪共计574头母猪的VEGF基因进行多态分析,并将基因多态性与猪总产仔数(TNB)和产活仔数(NBA)进行关联分析,旨在探讨猪VEGF基因多态性对其产仔数的影响。结果表明:在VEGF 5′调控区内有2个突变位点。P1产物第191位点处由A突变为G,在3个猪品种中只发现AA、AG 2种基因型,其中AG为优势基因型,A为优势等位基因。长白猪第1胎中AG型的TNB和NBA比AA型分别高出1.34头和1.44头(P<0.01),杜洛克猪所有胎次中AG与AA的TNB差异显著(P<0.05),NBA差异极显著(P<0.01)。P2产物第103位点处由C突变为T,在3个猪品种中发现TT、TC、CC 3种基因型,T为优势等位基因。TT、TC分别比CC型的TNB高出2.63头(P<0.05),在杜洛克所有胎次中TC比TT的TNB和NBA分别高出0.84头和0.94头(P<0.05)。P1位点和P2位点能否作为产仔性状的标记基因还需进一步研究。In order to find the effect of vascular endothelial growth factor(VEGF) polymorphism on litter size in pigs,the polymorphism of 5′regulatory region of VEGF gene was studied by PCR-SSCP in a total of 574 sows,including Landrace,Yorkshire,and Duroc,and the association between the polymorphism and litter size was analyzed by the least square means.The results indicated that there were two polymorphic loci within the 5′regulatory region of VEGF gene.At allele at P1 locus.The genotypes was very significantly(P0.01) associated with litter size in first parity in Landrace,and was significantly(P0.05) associated with litter size in all parities in Duroc.At P2 polymorphic locus,there was a C/T transition at position 103 bp of primer P2 PCR product,and there were three genotypes(TT,TC and CC),TC was superior genotype and T allele was superior allele.The genotypes was significantly(P0.05) associated with litter size in first parity and all parities in Landrace,and was significantly(P0.05) associated with litter size in the third to sixth parity and all parities in Duroc.The two locus as the litter size of marker genes need to be further researched.
