摘  要:目的:建立同时测定不同采收期青翘和连翘叶中连翘酯苷、芦丁和连翘苷含量的方法,确定青翘和连翘叶的最佳采收期。方法:采用高效液相色谱法。色谱柱为Alltima C18(250mm×4.6mm,5μm),流动相为甲醇-水(梯度洗脱),检测波长为280nm,柱温为室温,流速为0.8mL.min-1。结果:连翘酯苷、芦丁、连翘苷的进样量分别在0.74~14.80μg(r=0.9999)、0.163~3.260μg(r=0.9997)、0.293~5.860μg(r=0.9998)范围内与各自峰面积积分值呈良好线性关系;平均加样回收率分别为99.3%、97.6%、97.8%,RSD分别为2.48%、2.36%、2.66%(n均为6)。结论:本方法简便、快速、专属性强、灵敏度高、重复性好,可用于连翘的质量控制。青翘以7月份、连翘叶以6~7月份采收为宜。OBJECTIVE:To establish the method for the content determination of forsythiaside,rutin and phillyrin in the fruit and folium of Forsysia suspensa in different harvesting time and to confirm the optimal harvesting time of them.METHODS:HPLC method was adopted.The assay was performed on Alltima C18(250 mm×4.6 mm,5 μm) column with methanol-water as mobile phase(gradient elution) at a flow rate of 0.8 mL·min-1.The detection wavelength was 280 nm and column temperature was at room temperature.RESULTS:The linear ranges of forsythiaside,rutin and phillyrin were 0.74~14.80 μg(r=0.999 9),0.163~3.260 μg(r=0.999 7)and 0.293~5.860 μg(r=0.999 8),respectively.Average recoveries were 99.3%,97.6%,97.8% and RSDs were 2.48%,2.36% and 2.66%(n=6).CONCLUSION:The method is simple,rapid,specific,sensitive and reproducible for the quality control of F.suspensa.The suitable harvesting time for fruit of F.suspensa is in July,the suitable harvesting time for folium of F.suspensa is in June and July.
