摘 要:目的和方法:采用离体心房灌流模型,观察非特异性阿片受体拮抗剂纳络酮对离体心房的变时变力作用,旨在探讨纳络酮对心肌的直接作用。结果:①在15~120 μmol/L的浓度范围内,纳络酮呈剂量依赖性地降低右房的自发搏动频率,增加心房肌的收缩张力;②当α,β受体阻断或μ受体激动后,纳络酮对离体心房的作用仍然存在;③纳络酮可延长心房肌的功能性不应期。结论:纳络酮对心肌具有不依赖于中枢及交感神经系统的直接作用。Aim and Methods: To test the direct effects of naloxone (an non selective opioid antagonist) on heart, we employed the isolated atrial preparations to investigate the inotropic and chronotropic actions of naloxone. Results:①naloxone reduced the spontaneous rate and increased the contractile force in a concentration dependent manner ranging from 15 to 120μmol/L;②Neither α nor β adrenergic antagonist in dose that effectively block α and β receptor can block the action of naloxone, and the effects were not antagonized by pretreatment with morphine, either.③naloxone (120 μmol/L) could prolong the functional refractory period of atria. Conclusion: Naloxone may have direct effects on heart independent on central and sympathomedullary action. Heart may be another additional organ that respond to naloxone not through opiate antagonism.
