摘 要:目的观察条件恐惧应激条件下大鼠的行为及血液相关指标变化。方法 64只Wistar大鼠随机分为恐惧应激组及空白对照组,恐惧应激组按照改良Takeuchi法制作恐惧模型大鼠,空白对照组正常饲养。检测每周(共4周)体重、自发活动相关指标、血清皮质酮和超敏C反应蛋白(hs-CRP)浓度。结果恐惧应激组大鼠的冻结行为学得分高于空白对照组(t=5.700,P<0.01);恐惧应激组体重增长减慢(第一周t=0.457,P=0.642;第二周t=2.675,P<0.05;第三周t=2.316,P<0.05;第四周t=3.930,P<0.01);自发活动实验中恐惧应激组的活动次数减少(第一周t=2.267,P<0.05;第二周t=2.990,P<0.01;第三周t=3.325,P<0.01;第四周t=2.423,P<0.05),恐惧应激组的休息时间增多(第一周t=2.621,P<0.05;第二周t=3.356,P<0.01;第三周t=2.668,P<0.05;第四周t=2.384,P<0.05);恐惧应激组血清皮质酮浓度升高(第一周t=2.231,P<0.05;第二周t=3.531,P<0.01;第三周t=2.681,P<0.05;第四周t=2.225,P<0.05);恐惧应激组血清hs-CRP浓度升高(第一周t=2.773,P<0.05;第二周t=4.043,P<0.01;第三周t=3.373,P<0.01;第四周t=3.162,P<0.01)。结论应用Takeuchi法可建立条件恐惧模型,条件恐惧应激可使机体体重增长减慢,行为和神经内分泌系统发生变化。Objective To observe the effect of conditioned fear stress on mental behavior and serum corticosterone,C-reactive protein(CRP) in rats.Methods A total of 64 wistar rats were randomly divided into control group and conditioned fear stress group.The rats' model of stress was made by Takeuchi's method.The behavioral were recorded and analyzed,the weight,the level of the serum corticosterone and hs-CRP were determined to evaluate the model.Results Compared with the control group,behavioral score(t=5.700,P0.01),weight increase(week1:t=0.457,P=0.642;week2:t=2.675,P0.05;week3:t=2.316,P0.05;week4:t=3.930,P0.01),active times(week1:t=2.267,P0.05;week2:t=2.990,P0.01;week3:t=3.325,P0.01;week4:t=2.423,P0.05),rest time(week1:t=2.621,P0.05;week2:t=3.356,P0.01;week3:t=2.668,P0.05;week4:t=2.384,P0.05),level of the serum corticosterone(week1:t=2.231,P0.05;week2:t=3.531,P0.01;week3:t=2.681,P0.05;week4:t=2.225,P0.05) and hs-CRP(week1:t=2.773,P0.05;week2:t=4.043,P0.01;week3:t=3.373,P0.01;week4:t=3.162,P0.01) were differently significant in conditioned fear stress group.Conclusion Takeuchi's method can establish model of conditioned fear stress,conditioned fear stress can result in lower weight increase,influence behavior and neuroendocrine system in rats.
