摘  要:目的探讨热毒宁注射液体外抗肠道病毒(EV)的效果。方法采用细胞培养技术观察不同浓度热毒宁、炎琥宁、利巴韦林及不同给药方式下EV攻击后各组的细胞病变效应(CPE),并采用MTT法测定及蚀斑抑制实验,计算药物的TC50、TI及蚀斑抑制率。判断3种药物的抗病毒活性并进行前两者病毒吸附前后的药效比较。结果①热毒宁、炎琥宁、利巴韦林的TC50分别为52.0、2.0及2.0 mg/mL。热毒宁在1.3 mg/mL时即有抑制CBV3和ECHO11的活性,且与药物浓度成正相关;炎琥宁、利巴韦林分别在0.4~1.2 mg/mL及1.0~1.5 mg/mL之间有抑制CBV3和ECHO11的活性。②26.0 mg/mL热毒宁对CBV3及ECHO11蚀斑抑制率分别为79.8%及88.6%。0.8 mg/mL炎琥宁为67.0%及76.6%。1.5 mg/mL利巴韦林为44.2%及41.9%。③热毒宁在病毒吸附前用药蚀斑抑制率高于吸附后用药(P〈0.05),炎琥宁的差异则不显著(P〉0.05)。结论 3种药物均有体外抗CBV3及ECHO11的作用,但热毒宁的病毒抑制活性优于炎琥宁和利巴韦林,且毒性最小。热毒宁在病毒吸附前用药抗病毒活性优于病毒吸附后,有一定预防作用。【Objective】 To study the anti-enterovirus effect of reduning Inject in-vitro.【Methods】 Adopting the technique of cell culture to observe cytopathogenic effect(CPE) and MTT colorimetry,plaque-reduction assay to evaluate the anti-enterovirus effect of Reduning,Yanhuning and Ribavirin.The anti-enterovirus effect of Reduning and Yanhuning were also compared before and after ECHO 11 absorbing cytopathic plaque-reduction assay.【Results】 ①The TC50 of reduning,Yanhuning and ribavirin was 52.0 mg/mL,2.0 mg/mL,2.0 mg/mL respectively.Reduning could inhibit CBV3 and ECHO11 at the concentration of 1.3 mg/mL and the viral inhibiting effect was concentration-dependent.Yanhuning,ribavirin could inhibit CBV3 and ECHO11 at the concentration from 0.4 mg/mL to 1.2 mg/mL and from 0.4 mg/mL to 1.2 mg/mL respectively.②The plaque-reduction rate of 26.0 mg/mL reduning was 79.8% and 88.6%,of 0.8 mg/mL Yanhuning was 67.0% and 76.6%,of 1.5 mg/mL Ribavirin was 44.2% and 41.9% respectively.③The anti-ECHO11 activity of Reduning before ECHO11 absorbing was higher than after ECHO11 absorbing(P 0.05),there was no significant difference between the plaque-reduction rates of Yanhuning before and after ECHO11 absorbing(P 0.05).【Conclusion】 The three drugs all have the anti-virus activity in vitro while reduning are most effective.Among these three drugs,the cytotoxicity of reduning is most weak.The anti-virus activity of Reduning adding before virus absorbing was higher,so it seems that reduning can prevent EV infection.
