摘  要:比较了3种不同浓度的甘油(1.4,1.0,0.7mol/L)和投液氮温度(-25℃、-30℃和-35℃)对牛体外受精胚胎冷冻效果的影响。结果发现,最终投液氮温度为-25℃时,胚胎冻后存活率以1.4mol/L甘油组为最高,同1.0mol/L甘油组相比,差异显著(78.5%vs 64.5%,P<0.05);与0.7mol/L甘油组相比,差异极显著(78.5%vs53.5%,P<0.01)。以1.0mol/L的甘油冷冻,-30℃投液氮的效果优于-25℃的,它们之间冻后胚胎的孵化率差异显著(77.1%vs 64.5%,P<0.05)。用0.7mol/L甘油冷冻,投液氮温度以-30℃的为最好,冻后胚胎的孵化率(73.3%)极显著(P<0.01)高于-25℃的(53.5%)。用1.4mol/L甘油冷冻,以-25℃投液氮为最好,冻后胚胎的孵化率(78.5%)显著(P<0.05)高于-35℃的(65.7%)。结果表明,不同的甘油浓度和投液氮温度对牛体外受精胚胎的冷冻效果有显著的影响。This experiment was conducted to compare the survival rate of bovine IVF blastocysts frozen in different concentrations of glycerol(G) and temperatures before plunging into liquid nitrogen(LN2).The results showed that the hatching rate of embryos frozen in 1.4 mol/L G(78.5%) was significantly higher than that of embryos frozen in 1.0 mol/L G(64.5%,P0.05) or 0.7 mol/L G(53.5%,P0.01),while the day 7 blastocysts were plunged into LN2 at-25℃.The hatching rate of embryos frozen in 1.0 mol/L G or 0.7 mol/L G and plunged into LN2 at-30℃(77.1%,73.3%) were significantly higher(P0.05) than that of embryos plunged into LN2 at-25℃(64.5%,53.5%).When embryos were plunged into LN2 at-30℃ or-35℃,there was no significant difference in hatching rate among three glycerol concentrations groups.These results indicate that glycerol concentrations and temperatures before plunging into LN2 affects the viability of frozen-thawed embryos.
