摘 要:【目的】研究白木纳格葡萄败育型胚珠离体培养的最优方案,为木纳格葡萄胚挽救育种技术的研究奠定基础。【方法】以花后不同天数的白木纳格葡萄败育型胚珠为试材,进行离体培养,研究接种时间、基本培养基种类I、BA浓度以及活性炭对胚珠发育率的影响。【结果】白木纳格葡萄败育型胚珠取样适宜期为DAF35至DAF40,离体培养后,胚珠发育率均可达到50%以上,其中DAF 40 d为最佳取样时间,胚珠发育率可达到61.07%。在IBA 1.5 mg/L,GA30.5 mg/L,蔗糖60 g/L,琼脂5 g/L,活性炭1 g/L的Nitsch培养基中,白木纳格葡萄败育型胚珠发育率最高。【结论】建立了适合白木纳格葡萄败育型胚珠培养体系。【Objective】The purpose of this program was to research the optimal design of self-pollination of white Munake grape vestigial ovule that was used as test materials in this experiment,which would lay a theoretical basis of embryo rescue.【Method】The berries of white Munake grape were collected in different days after flower drop.The vestigal ovules were inoculated on medium used with various composition,and they were studied at the different sampling times.And the different basic medium,different concentration IBA and different concentration AC of vestigal ovules on the effect of culture were studied,too.【Result】The results showed that the best inoculation period of white Munake grape vestigal ovules was 35-40 days after flower drop stage,and in Nitsch medium supplemented IBA 1.5 mg/L,GA3 0.5 mg/L,suger 60 g/L,TC agar 5 g/L and AC 1 g/L,the percentage of ovules developed the highest.【Conclusion】This study provided the culture system of the vestigal ovule from white Munake grape.
