摘  要:目的研究青黛克银丸抗瘙痒的作用机制。方法建立瘙痒动物模型,观察青黛克银丸对实验动物内源性与外源性组胺释放的对抗作用;测定磷酸氯喹致痒小鼠血清中IL-2、IL-4的含量及计数抗血清致敏大鼠的颅骨骨膜肥大细胞脱颗粒,探讨青黛克银丸抗瘙痒的作用机制。结果青黛克银丸可显著减少4-AP致痒小鼠10 min的内瘙抓次数并明显缩短瘙痒持续时间,明显降低组胺模型小鼠腹部蓝斑面积与OD值并抑制大鼠颅骨骨膜肥大细胞脱颗粒。高、中剂量青黛克银丸可显著降低小鼠瘙痒后血清中IL-2的水平,而高剂量组可使血清中IL-4的含量明显升高。结论青黛克银丸抗瘙痒的作用机制可能与其抑制组胺释放和肥大细胞脱颗粒以及对IL-2、IL-4水平的调节作用有关。OBJECTIVE To study the mechanism of Qingdai Keyin pill on anti-pruritus.METHODS The antagonistic effects of Qingdai Keyin pill on endogenous and exogenous histamine release were observed in itching models.The serum content of IL-2 and IL-4 in mice with the body-scratching behavior caused by chloroquine phosphate was determined and rat's skull periost mast cell(MC) degranulation caused by antiserum was counted to discuss mechanism of Qingdaikeyin pill on anti-pruritus.RESULTS Qing-dai Keyin pill could clearly abate the frequency of scratching within 10 min and shorten scratching duration time in model mice induced by 4-AP,significantly decreace area of blue-stain skin and OD value in histamine model and inhibit the degranulation of MC in rats,The high and middle dose of Qingdai Keyin pill could decreace the levels of IL-2.Additionally the high dose of Qingdai Keyin pill could increase the content of IL-4 in mice serum.CONCLUSION Anti-pruritus effects mechanism of Qingdai Keyin pill may be attributed to the supperession of histamine release and MC degranulation in rats as well as the regulation effect for IL-2 and IL-4.
