摘 要:目的:研究长期、间断使用地塞米松(dexamethasone,Dex)对大鼠骨骼的影响。方法:20只雄性SD大鼠随机分为Dex组和NC组,Dex组肌注Dex,0.25 mg/100 g,NC组给予等体积的生理盐水,1周2次,共给药90 d。取胫骨分析皮质骨及松质骨的形态变化,股骨行骨生物力学检测。结果:Dex组皮质骨骨量明显低于NC组,骨髓腔增大(P<0.05);Dex降低骨内、外膜骨形成及矿化,但促进了骨内膜的骨吸收;Dex没有弱化股骨的力学性能。结论:长期、间断使用Dex,仅使皮质骨丢失,其几何形状的变化并不改变生物力学的性能,提示长期、间断使用Dex可以减轻该药对骨骼的不良影响。Objective: To study the long-term and intermittent use of the Dexamethasone (Dex) on rat bone. Methods: 20 cases of male SD rats were randomly divided into Dex group and normal control group (NC group). Dex group were given Dex by intramuscular, 0.25 mg/100g, NC group were given physiological saline with the same volume. Twice a week lasting for 90 days. The tibia was taken for bone histomorphometry, and the femur were taken for bone biomechanical experiment. Results: Dex was found to significantly reduce cortical bone and enlarge bone marrow cavity area compared with the NC group (P〈0.05). In periosteum and endosteal, Dex reduced the bone formation and mineralization. Moreover Dex enhanced endosteal bone resorption. In Dex treatment, the biomechanical properties of the femur were not weaken. Conclusion: The cortical bone is losen with long-term and intermittent use of Dex, but the biomechanical properties of cortical bone were not weakened with the change of geometrical shape. These results suggest that long-term and intermittent use of Dex may lessen the side effect of Dex on bone.
