摘 要:目的:建立微流控芯片非接触电导检测法测定盐酸吗啉胍片中盐酸吗啉胍含量。方法:在高电场强度条件下利用微流控芯片上特殊的微通道实现组分快速、高效的分离,以非接触电导检测器进行检测;探讨了缓冲液种类和浓度、添加剂种类和浓度、分离电压和进样时间等因素对分离检测的影响。结果:优化选择2 mmo·lL-1HAc+2 mmo·lL-1NaAc(pH=4.5)为缓冲溶液,0.3 mmo·lL-1十二烷基硫酸钠为添加剂,分离电压为2.20 kV,进样时间为10 s,2 m in内可实现盐酸吗啉胍快速分离和检测。在优化条件下,盐酸吗啉胍的线性范围为20~200μg.mL-1(r=0.9999),检出限为2.0μg.mL-1(S/N=3),RSD为1.4%,加标回收率为98.3%~101.2%。结论:方法简便快速,重复性好,适用于盐酸吗啉胍的快速检测和质量控制。Objective:To establish a new method for the content determination of moroxydine hydrochloride in moroxydine hydrochloride tablets by microfluidic chip with contactless conductivity detection.Methods:Moroxydine hydrochloride was rapidly separated by microfluidic chip and detected with contactless conductivity detection under high electric field.The electrophoretic parameters,such as the variety and concentration of buffer solution and additive,separation voltage and injection time,etc,were researched.Results:In optimal conditions,using 2 mmol·L-1 HAc+2 mmol·L-1 NaAc(pH=4.5) as buffer solution,with 0.3 mmol·L-1 SDS as additive at the separation voltage of 2.20 kV and 10 s injection,moroxydine hydrochloride was separated and detected within 2 min.The linear dependence of the concentration of moroxydine hydrochloride ranged from 20 μg·mL-1 to 200 μg·mL-1(r=0.9999),with the detection limit of 2.0 μg·mL-1(S/N=3),the RSD of 1.4% and the recoveries from 98.3% to 101.2%.Conclusion:The method is simple,rapid and reproducible,and it can be used for rapid detection and the quality control of the product.
