流式细胞仪在细胞凋亡检测中的应用

摘　　要：凋亡是一种细胞主动参与的自毁过程,与细胞坏死有根本区别。异常的凋亡状态与多种疾病密切相关,凋亡研究在临床有广泛的应用前景。本文介绍了4种应用流式细胞仪检测凋亡的方法。DNA含量分析最为常用,操作简便、成本低廉;Caspase-3是凋亡的早期检测指标,可以发现细胞内蛋白酶的升高;Annexin-Ⅴ探针能检测凋亡早期胞浆膜的改变;DNA断裂点标记是对凋亡形态学改变特征的流式检测方法。Apoptosis is used to describe a process in which a cell actively participates in its own destructive processes, which is quit different from 'necrosis . Inappropriate induction of apoptosis has been associatedwith many diseases and has broad ranging pathologic implications This article is intended to provide four flow cytometric approaches to identifying apoptosis ceil. DNA quantity analysis is a common and cost-effective method while the earliest observation is to identify and quantitative the activity of caspase -3, a key member of the caspase family of proteases Another earlier marker is to observe the redistribution of phosphotidylserine in plasma membrane by Annexin -V DNA fragment end-labeling is a characteristic and quantitative flow cytometric approach to identifying the apoptosis.
