摘 要:目的:探讨降钙素基因相关肽( CGRP) 基因转导兔动脉内膜对其舒缩功能的影响。方法:利用逆转录病毒载体,将CGRP 基因在体转导兔髂动脉内膜,取动脉环做灌流试验,研究其对血管活性物质的反应。结果:转基因血管内皮细胞的增殖较对照组活跃,动脉的内皮依赖性舒张反应较强,10 -6mol/L 的乙酰胆碱引起的舒张为33-08 % ±10-01 % ,对照组为- 17-57 % ±5-59 % ,P< 0-01 。结论:CGRP 基因转导损伤的免髂动脉内膜可促进内皮细胞增殖,并使动脉内皮依赖性舒张反应改善。AIM: To study the effect of calcitonin gene related peptide (CGRP) on contraction and relaxation of iliac artery transduced with CGRP gene by retrovirus vector. METHODS:Use retrovirus vector, the CGRP gene was transduced in the intima of the artery in vivo . Then the responses of artery to phenylephrine acetylcholine (Ach), sodium nitroprusside and norepinephrine were studied.RESULTS:Proliferation of endothelial cells in the artery transduced by CGRP gene was accelerated and endothelium dependent relaxation of the artery was improved, percentage of relaxation arisen by 10 6 mol/L Ach was 33.08%±10.00%,while in the control group it was -17 57%±5.59%, P <0.01 CONCLUSION:Transduction of CGRP gene can promote proliferation of endothelial cells and improve the function of endothelial cells in artery with injured intima.
