摘  要:目的:探讨白细胞与内皮细胞粘附对内皮单层通透性的影响。方法:将兔肺微血管内皮细胞单层培养在通透性测定装置内室底部的微孔滤膜上,然后与兔多形核白细胞共育,通过CD18 单抗封闭试验,观察缺氧时兔多形核白细胞与内皮细胞粘附率、[125I] - 白蛋白清除率、培养基中PAF 与NO 的变化及其相关关系。结果:缺氧时[125I] - 白蛋白清除率、培养液中PAF 与NO 水平与白细胞- 血管内皮细胞的粘附有显著相关性。结论:缺氧时内皮单层通透性部分依赖于白细胞- 血管内皮细胞的粘附,并与粘附所致的PAF、NO 释放有关。AIM: To investigate whether the polymorphonuclear leukocytes (PMN) adhering to rabbit pulmonary microvascular endothelial cells (PMVEC) may increase endothelial monolayer permeability(EMP) under hypoxia. METHODS: Confluent monolayer of PMVEC were grown on genatinized polycarbonate microporous filters at the bottom of a luminal chamber. The chamber was filled with medium in which rabbit PMN suspended. The luminal chamber was placed in an abluminal chamber filled with medium. Then the system was exposed to hypoxia (PO 2=15~20 mmHg). Adherence rate(AR),[ 125 I]-labled albumin clearance rate(CR), platelet activating factor(PAF), NO were assessed at different time points.RESULTS: Hypoxia resulted in a remarkably increasing of CR on comparing with normoxia group no matter whether PMN was added at all time points. And adding PMN to the system, CR increased evidently. The increased AR and CR by hypoxia can be reduced by McAbCD18 in a dose-dependent fashion. NO, PAF contents were also reduced. There was a positive correlation between the adherence rate and PAF and NO contents. CONCLUSION: During hypoxia, PMN adhering to endothelial cells increased EMP, it may be related to the release of PAF and NO induced by PMN adherence.
