银屑病患者骨髓造血干细胞中干扰素-γ的表达

摘　　要：目的探讨干扰素-γ(interferon,IFN-γ)在中重度寻常性银屑病患者骨髓造血干细胞中的表达情况,评价其与病情严重程度的相关性。方法免疫磁珠分选纯化20例中重度寻常性银屑病患者及10例健康对照者骨髓造血干细胞,以β-actin基因为内参采用Taqman探针实时定量PCR技术分析患者及健康对照者IFN-γ的mRNA水平,并进行统计学分析。结果 IFN-γ在寻常性银屑病患者及健康对照者的骨髓造血干细胞中均有表达,且在前者明显高表达,为后者的20.53倍(P<0.001);而IFN-γ的mRNA水平与银屑病的严重程度无相关性(r=0.014,P=0.631)。结论造血干细胞中IFN-γ的异常表达可能影响造血干细胞的增殖与分化,进而参与银屑病的发病。Objective To explore the abnormal expression of interferon（IFN）-γ in hematopoietic stem cells（HSCs） of patients with psoriasis,as well as the relations between IFN-γ mRNA level and severity of psoriasis.Methods HSCs,which was obtained from 20 patients with psoriasis and 10 healthy volunteers,were isolated and purified by magnetic bead.Real time polymerase chain reaction（PCR） technique was used to detect the mRNA level of IFN-γ,and then statistical analysis was performed.Results The level of IFN-γ mRNA in patients with psoriasis was 20.53-fold than that in control group（P0.001）.No correlation was found between IFN-γ mRNA level and severity of psoriasis（r=0.014,P=0.631）.Conclusion Abnormal expression of IFN-γ in HSCs may be an important mechanism for HSC growth regulation in psoriasis.
