消银解毒饮调控银屑病血热证肿瘤坏死因子-α的相关研究

摘　　要：目的研究消银解毒饮对银屑病血热证外周血单一核细胞(PBMCs)分泌肿瘤坏死因子-α(TNF-α)的调控作用。方法入选银屑病血热证患者抽取静脉血,分离PBMCs,并与人永生化角质形成细胞(HaCat细胞)混合接种培养48h,加入五种不同组别的动物药理血清,即蒸馏水空白组、消银解毒饮组、凉血方组、解毒方组、甲氨蝶呤阳性对照组,并于不同体积分数培养混合细胞48h,用四唑盐比色实验(MTT)测定药物对HaCat细胞的细胞增殖率,双抗体夹心法测定药物对混合培养细胞上清TNF-α的影响。结果消银解毒饮能有效的抑制HaCat细胞增殖(P<0.001),且效果优于拆方中药组(P<0.05);消银解毒饮能降低混合培养细胞分泌TNF-α(P<0.05),且与拆方两组比较疗效更显著(P<0.05)。结论消银解毒饮能有效的抑制角质细胞活化增殖,对银屑病血热证TNF-α有免疫调节作用,凉血类中药与解毒类中药协同作用治疗血热型银屑病效果可能更佳。Objective To study regulatory effects on Xiaoyin Jiedu decoction on tumor necrosis factor（TNF）-α production in psoriatic patients with blood-heat syndrome.Methods The venous blood was extracted.Then peripheral blood mononuclear cells（PBMCs） were separated and cultured with keratinocyte for 48 hours,at which time,different animal pharmacologic serum was added（five groups： water control group,Xiaoyin Jiedu decoction group,cool party group,blood detoxification party group,methotrexate positive control group）.After a 48-h our period of inoculation,proliferation rates of keratinocyte were measured by MTT,and the effect of medicine on cells secreting TNF-a was observed by double antibody sandwich assay method.Results Xiaoyin Jiedu decoction could controlled secretion of TNF-a in inoculated cells（P 0.05）,thereby inhibiting keratinocyte proliferation（P0.001）,and the effect was better than those of others.Conclusion Xiaoyin Jiedu decoction might regulate immunity of some cytokines such as TNF-a in certain cytokines in peripheral blood lymphocytes to suppress keratinocyte proliferation.
