摘　　要：目的:探讨蛋白酶体抑制剂硼替佐米对小鼠树突状细胞的免疫调节作用,从而促使其更加广泛应用于抗肿瘤治疗。方法:制备小鼠骨髓来源的树突状细胞,用CCK-8法检测硼替佐米对DC的抑制作用,流式细胞术检测DC表面共刺激分子的表达,ELISA法检测DC培养上清及MLR上清液中的细胞因子表达。结果:硼替佐米能够以时间、剂量依赖方式抑制DC的生长。结论:作为蛋白酶体抑制剂,硼替佐米是能够对小鼠树突状细胞产生一定的影响。Objective To investigate the immune regulation of proteasome inhibitor bortezomib on murine dendritic cells thus contribute more widely used anti-cancer therapy.Methods Mouse bone marrow-derived dendritic cells,CCK-8 was detected with bortezomib inhibition of DC,DC by flow cytometry the expression of costimulatory molecules,ELISA assay and MLR culture supernatant of DC the expression of cytokines in the supernatant.Results Bortezomib can be a time and dose dependent manner inhibited the growth of DC.Conclusion As a proteasome inhibitor,bortezomib was able to murine dendritic cells to produce certain effects.
