摘  要:目的探讨少突胶质细胞相关microRNA(m iRNA)对中枢神经系统神经前体细胞增殖的作用。方法利用D icer1 loxp/loxp基因型小鼠和Olig1-Cre品系小鼠杂交获得miRNA加工酶Dicer基因条件性敲除小鼠,再用免疫荧光染色和B rdU标记等技术检测这种Dicer条件性敲除小鼠大脑和小脑中神经前体细胞的增殖,以及脊髓中新生神经元的变化情况。结果 Dicer条件性敲除小鼠大脑中神经前体细胞增殖加快,而小脑中增殖细胞数目较正常小鼠减少(12%,P<0.05)。此外脊髓中神经元的数目并未受到影响。结论少突胶质细胞相关m iRNA对中枢神经系统的胶质细胞的发生和增殖具有时空特异性或差异性调节作用。Objective To investigate the effect of oligodendrocyte-related microRNAs(miRNAs) on the proliferation of neural progenitor cells in the central nerve system.Methods Heterozygous mice(Olig1-Cre) were crossed with mice(Dicer1loxp/loxp) to obtain crossbred mice(Olig1-Cre+/-;Dicer1loxp/wt),and the crossbred mice were then bred with mice(Dicer1loxp/loxp) to produce Dicer-CKO mice(Olig1-Cre+/-;Dicer1loxp/loxp).The Dicer-CKO mice were subject to immunohistochemical assay and BrdU labeling to evaluate the proliferation in oligodendrocyte precursor cells in the cerebrum and cerebellum and the change in the neuron number of the spinal cord.Results The proliferation of oligodendrocyte precursor cells was increased in the cerebral cortex but decreased in the cerebellum in the Dicer-CKO mice,while the number of neurons in the spinal cord had no change.Conclusion Oligodendrocyte-related miRNAs regulate the proliferation of neural progenitor cells differentially in the cerebrum and cerebellum.
