摘 要:探讨枸骨叶总皂苷分离纯化的最佳工艺条件及其油脂抗氧化作用,采用6因素5水平正交设计法考察洗脱液浓度、洗脱时间、pH值等因素对纯化的影响。用紫外分光光度法测定总皂苷的含量,计算吸附量、解吸率和枸骨总皂苷的含量,最后确定最佳工艺条件;通过碘量法检测枸骨叶总皂苷抗氧化活性。结果表明,AB-8树脂纯化枸骨叶总皂苷的最佳工艺为吸附时间5 h,药液pH值为4、洗脱液固比为10∶1、洗脱液pH值为5、洗脱液乙醇体积分数为80%、解吸振荡时间3 h。枸骨叶总皂苷对猪油具有较强的抗氧化活性,且具有剂量效应关系。In this paper, the optimal formula and conditions on separation and purification of saponins from Foliurn Ilicis Cornutae by macroporous resin and its antioxidative activity on oil were studied. An orthogonal experiment of 6 factors and 5 levels was used to study the effects on separation and purification including eluent concentration, elu- tion time, pH value, etc. The content of saponins in Folium llicis Cornutae was determined by ultraviolet spectrophotometry. Then, the adsorption capacity, the rate of desorption and the concentration of saponins were calculated to determine the suitable conditions. Antioxidation activity of saponins from Folium llicis Cornutae was studied by the io- dine method. The results showed that the optimized separation and purification technology of saponins from Folium llicis Cornutae by AB -8 was as follows:time of 5 h, solution pH value of 4, eluent - solid ratio of 10: 1, eluent pH value of 5, the concentration of 80% ethanol eluent, and desorption oscillation time of 3 h. So, saponins have strong antioxidation activity to lard and its activity is in proportion to saponins in dose.
