摘　　要：目的观察消痰散结方对裸鼠MKN-45人胃癌组织中血管内皮生长因子D(VEGF-D)表达的影响,探讨消痰散结方防治胃癌淋巴转移的作用机制。方法 30只裸鼠建立MKN-45人胃癌原位移植模型,并随机分为生理盐水组、5-氟尿嘧啶(5-Fu)组及消痰散结方组,每组10只,分别给予生理盐水、5-Fu和消痰散结方干预后,采用免疫组织化学和RT-PCR方法检测瘤组织VEGF-D蛋白及mRNA的表达。结果消痰散结方可不同程度下调肿瘤组织中VEGF-D蛋白和mRNA水平的表达,与生理盐水组比较,差异有统计学意义(P<0.05,P<0.01)。结论调控淋巴管生长因子VEGF-D蛋白和mRNA的表达是消痰散结方防治胃癌淋巴转移的可能机制之一。Objective To discover the mechanisms of Xiaotan Sanjie recipe in inhibiting gastric carcinoma lymphatic metastasis by observing expression of VEGF-D in gastric carcinoma orthotropic transplantation model.Methods Thirty mice were divided into saline group,5-Fu group and Xiaotan Sanjie recipe group randomly after the orthotopic transplantation model bearing human MKN-45 cell line was built.Each group had 10 nude mice,and was given NS,5-Fu and Xiaotan Sanjie recipe,respectively.After treatment,VEGF-D in the tumor tissue was stained by immunohistochemistry,and RT-PCR was applied to detect the expression of those molecules mRNA that encoded protein.Results Xiaotan Sanjie recipe reduced the expression of VEGF-D mRNA and encoding protein（P〈0.05,P〈0.01）,which affect the lymphatic metastasis of gastric carcinoma.Conclusion It might be one of the mechanisms of inhibiting lymphatic metastasis that Xiaotan Sanjie recipe can down-regulate the expression of VEGF-D protein and mRNA.
