摘  要:本研究应用PCR方法检测了中国3种旧大陆猴(恒河猴、食蟹猴及藏酋猴)的TRIMCyp嵌合基因型个体的存在状况,并首次发现携带TRIMCyp嵌合基因中国品系恒河猴。同时本实验应用建立RT-PCR方法在mRNA水平上进一步检测了恒河猴和食蟹猴的TRIMCyp嵌合基因序列和特性。其编码的融合蛋白TRIMCyp丧失了对HIV-1复制的限制功能,推测具有该基因型旧大陆猴对HIV-1更易感,可用于建立新型的艾滋病非人灵长类动物模型。本研究结果将促进逆转录病毒跨种间传播的研究和建立HIV-1易感非人灵长类动物模型。Based on recent publications, TRIMCyp, the fusion protein of TRIM5 a and CypA, lost its ability to restrict the replication of some retroviruses in the old world monkeys, such as human immunodeficiency virus (HIV-1). However, there are no reports on the distribution of TRIMCyp chimera gene in major old world monkeys, except Pig-tailed Macaque in China. In addition, several published investigations failed to find TRIMCyp gene in Chinese Rhesus Macaque. In this study, the existence of TR/MCyp, the TRIM5 gene with a retrotransposon CypA cDNA, in 3 major Chinese old world monkeys (Rhesus Macaque, Crab-eating Macaque and Thibetana Macaque) was investigated by PCR. For the first time, the TRIMCyp chimera gene was found existing in a small population of Chinese Rhesus Macaque. Further more, a RT-PCR protocol was developed to detect the mRNA of TRIMCyp in Rhesus Macaque and Crab-eating Macaque. Macaque with this chimera gene are considered susceptible to HIV-1since the TRIMCyp fusion protein does not effectively inhibit the replication of HIV-1. The TRIMCyp/TRIMCyp genotype Macaques have the potential to be developed as novel non-human primate (NHP) animal models for HIV researches. Our results will benefit the studies on cross-species transmission of retrovimses and the development of novel H1V-l-susceptible NHP models.
