摘  要:目的:探讨缺氧诱导因子(HIF-1α)、信号转导和转录活化因子3(STAT3)在大肠癌淋巴转移中的作用机制。方法:采用荧光实时定量聚合酶链反应和免疫组织化学染色方法检测60例大肠癌组织及远癌切端大肠癌组织HIF-1α、STAT3和血管内皮生长因子(VEGF)-C的mRNA转录水平和蛋白表达情况,同时以抗podoplanin单克隆抗体标记大肠癌微淋巴管计数淋巴管密度(LVD),分析上述指标与大肠癌淋巴结转移之间的相互关系。结果:大肠癌组织中HIF-1α、STAT3和VEGF-C分子的mRNA转录水平及蛋白表达水平明显高于远癌切端大肠癌组织(P<0.05),大肠癌组织中HIF-1α、STAT3与VEGF-C分子mRNA转录水平及蛋白表达水平呈正相关,有淋巴结转移患者LVD高于无淋巴结转移患者(P<0.05)。结论:大肠癌组织较远癌切端大肠组织有更为密集的淋巴系统,大肠癌组织中HIF-1α、STAT3、VEGF-C是调节大肠癌淋巴管形成的关键因子。Objective: To study the mechanism of hypoxia inducible factor-1α (HIF-1α), signal transducers and activator of transcription 3(STAT3) in lymphatic metastasis of colorectal carcinoma (CRC). Methods: The levels of HIF-1α, STAT3 and vascular endothelial growth factor (VEGF-C) mRNA transcription and protein expression of adenocarcinoma and surgical margin tissues were determined by real-time quantitative PCR using fluorescence and immunohistochemistry in 60 cases with CRC. The lymph vessel density was counted using anti-podoplanin monoclonal antibody. And correlations of above indexes and lymphatic metastasis of colorectal carcinoma were analyzed. Results: The levels of HIF-1α, STAT3 and VEGF-C were significantly higher in tumor tissues than those of surgical margin in either mRNA transcription or protein expression (P 0.05). The levels of HIF-1α, STAT3 and VEGF-Cin mRNA transcription were positively related with protein expression of tumor tissues. But the levels were higher in lymphatic metastasis group compared with those without lymphatic metastasis group (P 0.05). Conclusion: The expression of lymphatic vessels system is higher in colorectal carcinoma than that in surgical marginal tissue. HIF-1α, STAT3 and VEGF-C are the key regulators in lymphatic vessel formation of colorectal carcinoma.
