摘  要:目的应用RNA干扰(RNAi)技术沉默Ki67基因,探讨其对裸鼠人肾癌细胞移植瘤细胞生长及凋亡的影响。方法建立裸鼠人肾癌细胞移植瘤模型,随机分组,于肿瘤内分别注射生理盐水0.2 ml(C组)、pSilencer空质粒20μg/只(P组)、pSilencer-Ki67重组质粒20μg/只(PK组),每隔2天注射一次,共7次。取肿瘤测量体积,免疫组化染色检测Ki67的表达,TUNEL法检测细胞凋亡。结果 PK组移植瘤体积明显小于C、P组,移植瘤细胞Ki67表达率也明显降低,肿瘤细胞凋亡率明显增加(P<0.01)。结论应用RNAi技术沉默Ki67基因可以降低人肾癌细胞裸鼠移植瘤Ki67基因表达,同时引起细胞凋亡进而抑制肿瘤的生长。Objective To investigate the effects of silence Ki67 gene with RNAi on the growth and apoptosis of human renal carcinoma xenografts in nude mice.Methods After establishment of nude mice model with human renal carcinoma xenografts,the xenograft was injected normal saline 0.2ml(group C),empty plasmid silencer 20μg(group P) or plasmid silencer with Ki67 20μg(group PK) every other two days for 7 times,respectively.The tumors were taken out for measuring the size,detecting the expression of Ki67 protein with immunohistochemistry analysis,and evaluating apoptosis by TUNEL assay.Results The size of xenograft was smaller,Ki67 positive expression rate was lower,and the apoptosis rate was higher in group PK than those in groups of C and P(P〈0.01).Conclusion Silencing Ki67 gene can decrease Ki67 gene expressions,induce cell apoptosis and suppress tumor growth in the nude mice model with human renal carcinoma xenografts.
