摘 要:目的探讨转化生长因子岛(TGF-β1)和Ⅰ、Ⅲ型胶原蛋白(COL-Ⅰ、COL-Ⅲ)在周围神经慢性卡压中的作用及二者的相关性。方法将50只成年雄性SD大鼠随机分成2组:对照组,仅分离暴露坐骨神经;卡压组,对大鼠右侧后腿坐骨神经行硅胶管卡压术。于卡压术后2.4、6、8、10周随机取2组大鼠各5只,切取卡压段神经行组织形态学、免疫组织化学观察及RT-PCR、WesternBlot定量测定TGF-β1及COL-Ⅰ、COL-Ⅲ含量。结果坐骨神经慢性卡压损伤后,神经内胶原纤维增生;卡压组大鼠坐骨神经TGF-β1及COL-I、COL-Ⅲ含量明显高于对照组,两者差异有统计学意义(P〈0.05);TGF-β1与COL-Ⅰ、COL-Ⅲ含量在一定时期内呈正相关。结论慢性卡压损伤可致周围神经发生纤维化病变,COL-Ⅰ、COL-Ⅲ含量显著增加,TGF-β1参与了这一病理生理改变过程,提示TGF-β1在周围神经纤维化过程中具有一定的作用。Objective To investigate the effects of transforming growth factor β1(TGF-β1) and collagen type Ⅰ/Ⅲ (COL- Ⅰ/Ⅲ ) on chronic peripheral nerve compression injury and explore the relationship between TGF-β1 and COL- Ⅰ/Ⅲ. Methods Fifty adult male SD rats were randomly divided into group A and B. In group A (sham-operated group) the sciatic nerve was exposed only. In group B (compression group) sciatic nerve entrapment was created on the right hind leg according to Mackinnon' s procedure of chronic sciatic nerve compression. Electron microscopy, inrmunohistechemistry, RT-PCR and Western-Blot were performed to observe the morphological changes of the compressed nerve tissue and to determine the level of TGF-~, collagen- Ⅰ and collagen-Ⅲ 2, 4, 6, 8 and 10 weeks after the surgery, respectively. Results After sciatic nerve compression, collagen content in the nerve increased. The expression of TGF-β1 and COL- Ⅰ/Ⅲ in the sciatic nerve of the compressed group increased, which was significantly higher than that of the sham-operation group ( P 〈 0.05). In the meanwhile, the contents of TGF-β1 and COL- Ⅰ/Ⅲ were positively correlated in a certain period. Conclusion Peripheral nerve fibrosis can he caused by chronic nerve compression which induces increased expression of COL-Ⅰ/Ⅲ in the nerve. TGF-I~ is involved in the pathophysiolagical process, which suggests that TGF-β1 plays an important role in the process of neural injury and fibrosis.
