肝细胞生长因子协同作用的研究

摘　　要：目的；探讨乳猪肝源性与肠源性ＨＧＦ活性物质联合应用在促进肝细胞再生方面的作用。：应用「＾３Ｈ」－ＨｄＲ掺入法观察ＨＧＦ对各实验组体外原代肝细胞培养及小鼠活体肝细胞刺激作用。结果；乳猪小肠粘膜存在ＨＧＦ活性而且与肝源性ＨＧＦ有明显的协同作用，同时证明这种协同作用并非胰蛋白酶所致。结论：乳猪肝源性与肠源性ＨＧＦ活性物质在刺激肝细胞再生方面具有协同作用，可作为ＨＧＦ制备过程中综合利用乳猪原材料的依据。Objective:To explore the possible synergetic effect of hepatocyte growth factor (HGC) from the liver and intestinal mucous of new-born pig on stimulating the proliferation of hepatocytes.Methods:The stimulating effect was determined with [~2H]dTdR incorporated into primary culture human hepatocytes DNA in vitro and rats liver cells DNA in vivo.Result:Compared with control group,the intestinal mucous has HGF activity (P<0.05),and it has significant synergetic stimulating effect with HGF from the intestine and liver (P<0.05).Trypsin inhibitor didn't change this synergism.Conclusion:The HGF from intestinal mucous has the activity of stimulating hepaocyte proliferation.It is obvious that there is a synergetic effect of HGF from liver and intestinal mucous and the effect ins't related to trypsin.It may be an evidence of comprehensive utilization for the new-born pig's raw metarial to prepare HGF.
