摘  要:目的:寻找一种快速有效地分离体外制备糖基化终产物(AGEs) 的方法。方法:通过葡萄糖与牛血清白蛋白(BSA) 共同孵育体外制备AGEs- BSA, 建立快速灌注色谱法并优化各种参数,比较不同程度及药物干预糖基化反应后的分离结果。结果:经优化相关参数建立的快速灌注色谱法能满意地进行糖基化反应体系中AGEs- BSA和BSA的分析性和制备性分离,AGEs- BSA的峰高和峰下面积可作为糖基化程度的间接指标。Objective:The experiment was designed to explore and establish a suitable and fast method for purifying advanced glycosylation end products(AGEs) prepared in vitro . Method:AGEs-BSA were prepared by incubating bovine serum albumin (BSA) with glucose. For analytic and preparative purification, perfusion chromatography was applied to isolate glycosylated and unmodified BSA after parameters of chromatography were optimized. Results:AGEs-BSA and unmodified BSA could be separated as two peaks without any overlap after optimization of the method. Both analytic and preparative isolation of AGEs-BSA could be achieved effectively by perfusion chromatography. Conclusion: The perfusion chromatography is thought to be a satisfactory and fast method for purification of AGEs.
