摘 要:目的应用含通脉丸的培养基进行实验研究,探讨其对人内皮祖细胞(endothelial progenitor cells,EPCs)功能的影响。方法采用密度梯度离心法分离出人外周血EPCs,实验分为正常对照组、低剂量组(10mg/ml)、中剂量组(20mg/ml)和高剂量组(50mg/ml),培养细胞7d。通过倒置显微镜、MTT实验以及细胞黏附、迁移实验评价EPCs增殖、黏附和迁移能力;采用细胞表型分析、VEGF的表达、NO的分泌和体外成血管能力来检测不同组别对细胞功能的影响。结果与对照组比较,不同浓度药物组能提高细胞的增殖、分化、迁移和黏附能力,并促进EPCs表型的改变、NO的分泌和VEGF的表达,形成丰富的血管样结构,差异有统计学意义(P<0.05,P<0.01);与低、高浓度组比较,中浓度组在细胞的增殖、黏附、迁移功能以及CD133表达和NO分泌等方面作用最为明显,差异有统计学意义(P<0.05)。结论通脉丸能明显改善外周血EPCs的功能,促使EPCs的增殖、分化、黏附和迁移,促进EPCs分泌NO和VEPF的表达,能够更快形成血管样结构。Objective Culture medium containing liquid medicine of "Tongmai Pill"(TMP) was used to explore the effects of TMP on the function of human endothelial progenitor cells(EPCs).Methods EPCs were isolated from human peripheral blood by density gradient centrifugation.Cultured EPCs were successfully identified after culture.The cultured cells were randomized into 4 groups: control group,low-dose group(10mg/ml),middle-dose group(20mg/ml) and high-dose group(50mg/ml) of TMP.After being cultured in EBM-2 containing different concentrations of TMP for 7 days,the invert microscope,MTT assay,cell adhesion and migration test were used to evaluate cellulate capability of proliferation,adhesion and migration.In addition,the effects in different groups on the function of cell were assessed by phenotype analysis from the aspects of NO production,VEGF expression and capillary tube formation.Results EPCs proliferative,migratory and adhesive capacities were remarkably improved by TMP.Furthermore,low-,middle-and high-dose groups could improve NO secretion,VEGF expression and capillary tube formation(P0.05 or P0.01);while middle-dose group of TMP was more prominent to improve proliferative,migratory and adhesive capacities,CD133 expression and NO secretion than low-dose group and high-dose group(P0.05).Conclusion TMP can significantly improve the function of human vascular endothelial progenitor cells.
