摘  要:目的:研究抑肽酶对心肌缺血再灌注损伤的保护作用。方法:SD 大鼠24 只,随机分为3组,每组8 只,采用离体鼠心非工作模型。A、B两组为实验组,灌注液分别含抑肽酶1×106 kIU·L1 和1 ×105 kIU·L1 ;C组为对照组,灌注液中不含抑肽酶。记录稳定期、药物灌注期和再灌注期的心功能、冠脉流量,再灌注第1 分钟测定心肌酶水平,第10 分钟测定氧耗量,实验后制电镜标本并测心肌湿/ 干质量比。结果:(1)在稳定期和药物灌注期,3 组心脏的心功能指标无显著性差异;(2) 再灌注期,A组鼠心的功能恢复、冠脉流量、心肌酶水平、心肌超微结构等方面指标明显优于B、C组。Objective:The protective effect of aprotinin on ischemia and reperfused myocardium was studied using isolated non-working rat hearts.Methods:24 SD rats were randomly divided into three groups of 8 animals in each.In experiment groups A and B,aprotinin 1×10 6 kIU·L 1 and 1×10 5 kIU·L 1 was added into K-H solution,in control group C no aprotinin was added.The heart rate,left ventricular diastolic pressure,±d p /d t max and coronary flow(CF) were monitored in the stabilization,perfusion and reperfusion period.Creatine phosphokinase(CPK) and lactate dehydrogenase(LDH) activity levels were measured in fluent at the first minute.Oxygen consumed(OC) was measured at the tenth minute after the start of reperfusion.Myocardial ultrastructure,wet/dry weight ratio of myocardium were examined after experiment.Results:1.The cardiac functions of three groups have no statistically significant difference during stabilization and drug perfusion period.2.Recovery of cardiac functions,CF,OC,CPK and LDH activity levels,wet/dry weigh ratio of the myocardium and myocardial ultrastructure in group A were much better than those of groups B,C during reperfusion.Conclusion:Aprotinin at a higher dose has protective effects on ischemia and reperfusion myocardium injury.
