摘 要:目的:了解谷胱甘肽(GSH)对糖基化终末产物(AGE)环境中人脐静脉内皮细胞(HUVEC)前列环素(PGI2)和内皮素-1(ET-1)分泌的影响。方法:分别用含M199培养基、含AGE(0.8g/L)的M199培养基、含GSH(6mmol/L)的M199培养基及含AGE(0.8g/L)和GSH(6mmol/L)的M199培养基培养HUVEC,培养24、48、72h后测定各组细胞中PGI2和ET-1的分泌量。结果:AGE可显著降低HUVEC的PGI2分泌量(24、48、72h时的FAGE=61.581,142.071和98.203,P均<0.001),升高ET-1分泌量(24、48、72h时的FAGE=22.927,63.229和26.962,P均<0.001);GSH可拮抗上述作用[(PGI2:48、72h时的F交互=8.121和6.857,P均<0.05;ET-1:48、72h时的F交互=105.892和49.257,P均<0.001)]。结论:GSH可减轻AGE对血管内皮细胞的损伤。Aim:To investigate the influence of reduced glutathione(GSH)on prostacyclin(PGI2)and endothelin-1(ET-1)secretion of human umbilical vein endothelial cells(HUVEC)cultured with advanced glycation end product(AGE).Methods:The secretion of PGI2 and ET-1 of HUVEC,which were cultured in M199 medium with AGE(0.8 g/L),with GSH(6 mmol/L),with AGE(0.8 g/L)and GSH(6 mmol/L),and M199 alone for 24,48,and 72 h,respectively,was assayed by RIA.Results:AGE could decrease the secretion of PGI2 from HUVEC(at 24,48,and 72 h,FAGE=61.581,142.071 and 98.203,P0.001),increase the secretion of ET-1(at 24,48,and 72 h,FAGE=22.927,63.229 and 26.962,P0.001).The decrease and increase could be diminished by GSH(PGI2:at 48 and 72 h,Finteraction=8.129,6.857,and 6.857,P0.05;ET-1:at 48 and 72 h,Finteraction=105.892 and 48.258,P0.001).Conclusion:GSH could protect vascular endothelial cells from the injury induced by AGE.
