摘  要:为探讨中药槐米中可溶性粗多糖的提取工艺,通过单因素试验和L9(33)正交试验,研究了提取温度、料水质量比、提取时间对多糖提取率的影响。结果显示,3个因素对多糖提取率的影响顺序为提取时间、料液比、提取温度,最佳提取工艺为提取温度90℃,料水质量比1∶15,提取时间90 min,所得槐米粗多糖的提取率为5.10%。对槐米粗多糖脱蛋白方法的研究显示木瓜蛋白酶+Sevag法脱蛋白处理效果优于Sevag法和3%三氯乙酸法。Single factor test and orthogonal design of L9(33) were adopted to analyze the extracting technology of polysaccharide from Flos Sophorae Immaturus.The results indicated that among the factors influencing the extracting rate,extracting time had the greatest impact,followed by solid to liquid ratio,while extracting temperature had the least impact.The optimum extracting technology was temperature 90 ℃,solid to liquid ratio 1∶15(m/V,g∶mL);and extracting time 90 min.Under these conditions,the extracting ratio of soluble polysaccharide from Flos Sophorae Immaturus was 5.10%.In addition,the study on protein removing method from crude polysaccharide revealed that Sevag method combined with papain was better than Sevag method alone or 3% TCA method.
