摘 要:目的研究动脉粥样硬化兔在右锁骨下动脉盗血模型建立下大脑后分水岭区皮层中MMP-9和TIMP-1的动态表达变化情况。方法 30只动脉粥样硬化兔,随机分为对照组、盗血后1d组和盗血后3d组,每组10只;取大脑后分水岭区皮层,采用荧光定量PCR技术检测MMP-9和TIMP-1的基因表达,并进行比较。结果 MMP-9在盗血后1d及3d上升,但组间未见明显统计学差异;TIMP-1表达在对照组最低,盗血后1d表达升高,在盗血后3d TIMP-1表达开始下降,但仍高于对照组。统计学处理后未发现组间有明显统计学差异。结论在该动脉粥样硬化兔右锁骨下动脉盗血模型中,分水岭区皮层中MMP-9和TIMP-1的基因表达在模型建立后的1d和3d呈现血管重建启动的变化趋势,但差异无统计学意义。该动物模型引起MMP-9和TIMP-1的表达变化情况需要进一步实验证实。Objective To investigate the dynamic changes of MMP-9 and TIMP-1 in the cortex of the brain watershed area of the atherosclerosis rabbit of right subclavian atery steal model. Methods Thirty healthy male New Zealand white rabbits were randomly assigned to control group, one-day steal model group and three-day steal group (n=10). With those killed and brain extracted the expressions of the gene of MMP-9 and TIMP-1 were measured by the technique of immunofluorescence PCR, and the differences among groups were analysed. Results As statistically showed, MMP-9 expression was a little higher in the three-day steal group than that in the one-day group, with both higher than that in the control group, although there was no significant difference among groups.TIMP-1 expression was the lowest in the control group, raised in the one-day steal group and began descending in the three-day steal group, but still higher than that in the control group. There was still no significant difference among groups. Conclusion In the right subclavian steal model of atherosclerosis rabbit, the gene and protein expression of MMP-9 and TIMP-1 in the cortex of the brain watershed area in the ond-day steal group and three-day steal group take on a variation trend of vessel reconstruction start, while there is still no significant difference among groups. Whether this model can give rise to the change of the expressions of MMP-9 and TIMP-1 needs to be further proved through experiments.
