摘 要:目的探讨眼针对脑缺血再灌注损伤(CIRI)模型大鼠脑组织中TLR4蛋白表达的影响及作用机制。方法健康雄性SPF级Wistar大鼠60只,随机分为正常组、假手术组、CIRI模型组和眼针组。CIRI模型组和眼针组采用改良的线栓法建立大鼠大脑中动脉缺血(MACO)再灌注动物模型。RT-PCR及免疫组化方法检测大鼠缺血侧脑皮层TLR4 mRNA和蛋白表达的变化。结果与正常组及假手术组比较,模型组大鼠缺血侧脑皮层TLR4 mRNA和蛋白水平均显示出高表达(P<0.01),眼针组与模型组相比,TLR4 mRNA和蛋白表达则下调(P<0.05)。结论眼针抑制脑皮层组织中TLR4的表达,可能是眼针治疗CIRI的主要作用机制之一。Objective To evaluate effects of eye acupuncture on the expression of TLR4 protein in rat's brain cortex after cerebral ischemia-reperfusion injury(CIRI) and probe into the mechanisms.Methods Adult male SPF Wistar rats were randomly divided into normal,sham operation,CIRI model and eye acupuncture groups,15 rats in every group.The model of middle cerebral artery occlusion(MACO) was established by the improved suture method.RT-PCR and immunohistochemical method were performed to examine the mRNA and protein expression of TLR4 in the ischemic brain cortex.Results The mRNA and protein expression of TLR4 in CIRI model group were increased significantly(P0.01) compared with those in normal and sham operation groups,and which were decreased significantly(P0.05) than those in eye acupuncture group.Conclusions Eye acupuncture therapy can reduce the expression of TLR4 in CIRI,which may be the major mechanism for CIRI treatment.
