摘 要:目的:运用基因芯片技术,研究激素性股骨头坏死骨代谢相关基因的差异表达情况,进一步探讨其骨质疏松学说发病机理的分子机制。方法:Wistar大鼠20只,随机分为对照组(n=10)和模型组(n=10),利用内毒素和甲强龙诱导激素性股骨头坏死模型,提取股骨头组织总RNA,采用基因芯片检测骨代谢关基因的表达改变。结果:基因芯片筛选出模型组与对照组大鼠与骨代谢相关的差异表达基因9条(Ratio>2.0)。结论:在激素性股骨头坏死的过程中,骨代谢相关基因的表达受抑制,进而影响骨重塑和软骨代谢,导致骨质疏松。Objective: To study the expression differences of the bone metabolism-related gene in the steroid-induced osteonecrosis by using gene chip technology,to further explore the theory of the molecular mechanism of the osteoporosis pathogenesis.Methods: 20 Wistar rats were randomly divided into control group(n=10) and model group(n=10).Endotoxin and methylprednisolone were used to induce model of femoral head necrosis,the total RNA of femoral head tissue were extracted,to test the expression changes of the bone metabolism-related gene by using DNA microarray.Results: There were nine differentially expressed genes related to bone metabolism between the control group and the model group.(Ratio 2.0).Conclusion: In the process of steroid-induced osteonecrosis,the expression of bone metabolism genes is down-regulated,thereby affecting bone remodeling and cartilage metabolism,leading to osteoporosis.
