摘 要:目的探讨内皮素-1反义脱氧寡核苷酸(ET-1AS-ODN)对链脲佐菌素(streptozocin,STZ)诱导的糖尿病大鼠心肌重构效应的保护作用及其机制。方法一次性腹腔注射STZ(60mg.kg-1),建立大鼠糖尿病模型,并观察ET-1AS-ODN预防性给药对大鼠糖尿病心肌重构效应的保护作用。结果实验动物饲养5周后,与生理盐水对照(NC)组相比,糖尿病模型组大鼠体重明显减轻(P<0.05),心肌细胞重构明显,表明糖尿病心肌病变(diabetic cardiomyopathy,DC)模型已构建;与DC组比较,ET-1AS-ODN各治疗组心脏指数和心室指数均减小;大鼠心肌VG染色观察仅见心肌细胞间有少量胶原纤维沉积,但大部分心肌纤维结构完整,横纹清楚。结论内皮素-1反义寡核苷酸可减轻STZ诱导的糖尿病大鼠的心肌重构效应,对心肌组织具有保护作用。其机制可能与ET-1 AS-ODN降低糖尿病大鼠内源性ET水平,减少胶原合成,从而改善心肌重构效应有关。Objective To investigate the Protective effects of endothelin-1 mRNA antisense oligodeoxynucleotide(ET-1AS-ODN) on myocardial remodeling and its mechanism in streptozotocin-induced diabetic rats.Methods Diabetic rats were induced by intraperitoneal administration of streptozotocin(STZ) at 60mg/kg.ET-1AS-ODN was administrated intravenously to the treatment groups and its protective effects on myocardial remodeling of diabetic cardiomyopathy were observed.Results Compared with the normal control,the body weight of the DM group decreased significantly(P0.05) with increased cardiac index and left ventricle index.Compared with the DM group,cardiac index and ventricular index were reduced in ET-1 AS-ODN groups(P0.05).VG dying showed the myoarchitecture was intact with slight collagen deposits in the myocardial tissue.Conclusion ET-1 AS-ODN can attenuate the effect of myocardium remodeling and protect the heart of the DC rats.The mechanism of its protective effect may involve in inhibition of endogenous ET-1 production and diminution of cardiac fibrosis in diabetes.
