宁夏回汉族人群MBL基因G54A位点多态性的分布特征

摘　　要：目的探讨甘露聚糖结合凝集素(MBL)结构基因外显子1第54位密码子点突变G54A多态性在宁夏回汉族人群的分布特征。方法应用聚合酶链式反应-限制性片段长度多态性(PCR-RFLP)技术检测MBL基因外显子1第54位密码子点突变G54A的基因型。结果 (1)宁夏回与汉族群体的MBL基因54(G/A)多态性位点基因型频率和等位基因频率的分布差异无统计学意义(P>0.05);(2)宁夏群体的基因型频率分布与高加索人等群体差异无统计学意义(P>0.05),而与广东汉族群体以及日本群体比较差异有统计学意义(P<0.01);(3)宁夏人群第54位密码子等位基因频率与高加索人等群体差异无统计学意义(P>0.05),而等位基因频率与广东汉族、张家口汉族以及日本群体差异有统计学意义(P<0.01)。结论不同的区域种族群体MBL结构基因外显子1第54位密码子的基因型频率和等位基因频率分布不尽一致。Objective To detect the point mutation of exon 1,codon 54 of Mannan binding lectin（MBL） in healthy Han and Hui population of Ningxia.Methods The mutated G54A of MBL exon 1 codon 54 genotype was detected by Polymerase chain reaction-restriction fragment length polymorphisms（PCR-RFLP） techniques.Results（1） MBL gene 54（G/A） polymorphism genotype distribution and allele frequencies were not statistically different between Ningxia Han and Hui population,（P0.05）;（2） Compared with the Guangdong Han population,the mutation genotype frequency in Ningxia groups were significant higher（P0.01）,but there was no significant difference compare with Caucasian and other groups（P0.05）.（3） Allele frequency of 54 codon of MBL showed significantly different in Ningxia population compared with the Guangdong,zhangjiakou Han population and Japanese（P0.01）,while there was no significant difference compare with Caucasian and other groups（P0.05）.Conclusion The genotype and allele frequency distributions of MBL exon 1 codon 54 in different region and ethnic groups are not consistent.
