摘 要:对香雪兰愈伤组织超低温保存的几个因素进行了初步的讨论.实验结果表明:香雪兰愈伤组织的适宜冷冻年龄为15~20d较佳的冷冻程序是从0℃直接投入液氮(-169℃)的快速冷冻法;氯化三苯四氮唑(TTC)在4℃化冻和40℃水浴化冻时有着较高的光密度值.In this experiment, everal factors about calli cryopreservation of Freesia refracta Klatt. were studied, for providing essential foundations to go deep into next experiments. The results showed that the 15th to 20th day calli had higher TTC values, and so they were suitable materials used as cryopreservation. The better freezing procedure was to reduce the temperature from 0℃ to - 196℃ ,which was called as rapid - freezing method. Thaw at 4℃ and warm - water bath at 40℃ had better effects than flowing - water wash and thawing under room temperature.
