摘 要:目的观察短期应用常用利尿剂呋塞米、双氢克尿噻及安体舒通对正常大鼠尿液水通道蛋白-2(aquaporin-2)排泄浓度的影响。方法将30只健康SD大鼠随机分成4组:呋塞米组、安体舒通组、双氢克尿噻组和对照组。观测尿量、及尿渗量变化情况,应用酶联免疫吸附测定法(ELISA法)检测尿液AQP2浓度。结果用药前后比较,各利尿剂组大鼠尿量均显著增多(P<0.05),尿液中的AQP2浓度和排泄率均明显增加(P<0.05)。但呋塞米组尿渗量低于对照组(P<0.05),而双氢克尿噻组尿渗量高于对照组(P<0.05)。结论 3类利尿剂均可显著增加正常大鼠尿液AQP2蛋白的排泄。Objective To investigate effects of the furosemide,antisterone and dihydrocholorothiazide on expression of kidney aquaporin-2 gene and urine aquaporin-2 excretion in rats in a short time.Methods Thirty Sprague dawley(SD) rats were randomized into 3 groups,namely the furosemide group,antisterone group and dihydrocholorothiazide group respectively.The urime were collected for detecting urine volume and urine osmolality during medicine treatment.Urine AQP2 concentration was measured by enzyme-linked immunosobentassay(ELISA).Results Urine volume and urinary AQP2 excretion were increased in rats of three treatment groups as compared with the begining.However,Urine osmolality was lower in rats of furosemide than the beginning.However,it was higher in dihydrocholorothiazide group as compered with the begining(P0.05).Conclusion Three classes of diuretics could increase the excretion of the urinary AQP2 in normal rats.
