滋阴清热方对缺乏淋巴增殖基因狼疮鼠CD4^+CD25^+T细胞亚群及Foxp3表达的影响

摘　　要：目的:探讨滋阴清热方对缺乏淋巴增殖基因狼疮鼠CD4+CD25+T细胞亚群及Foxp3表达的影响。方法:选用自发性系统性红斑狼疮(SLE)模型MRL/lpr小鼠40只,随机平均分为4组,即模型组、滋阴狼疮方治疗组(中药组)、强的松治疗组(西药组)、滋阴清热复方+强的松治疗组(中西药组)。另选用昆明小鼠10只作为对照组。连续用药10周后,流式细胞仪分析外周血CD4+CD25+T细胞亚群的百分比及CD4+CD25highT细胞与CD4+CD25intT细胞的比率,RT-PCR检测T细胞Foxp3 mRNA的表达。结果:各组外周血CD4+CD25+T细胞、CD4+CD25highT细胞和CD4+CD25intT细胞占CD4+T细胞的百分比组间比较,无统计学差异(P>0.05),而CD4+CD25highT细胞与CD4+CD25intT细胞的比率和T细胞Foxp3 mRNA表达水平,模型组与对照组比较,明显降低,差异有统计学意义(P<0.05),各治疗组均高于模型组(P<0.05),尤其是中西医结合组(P<0.01)。结论:中药滋阴狼疮方可上调MRL/lpr狼疮鼠Foxp3 mRNA表达水平,从而调节CD4+CD25+T细胞亚群比率,发挥对SLE的免疫治疗作用。Objective：To explore the influence of Compound Ziyin Qingre Prescri-ption on CD4＋CD25＋T Cell Subpopulations and Foxp3 gene of MRL/lpr Lupus Mice.Methods：40 MRL/lpr mice were randomly divided into four groups,model control group,TCM group,West medicine group and TCM and West medicine group,10 mice as control group.Then intragastrically administrated normal sodium,Compound Ziyin Qingre Prescription Decoction,prednisone suspension and Compound Ziyin Qingre Prescription Decoction plus prednisone suspension,0.5mL every time,once daily for 10 weeks respectively.Meanwhile,10 Kungming mice were devided into normal group,administrated normal sodium,like the model group.The peripheral blood mononuclear cells（PBMC） from these mice were stained for CD3,CD4 and CD25.Cells were examined by flow eytometry.The expression of Foxp3 gene were examined by RT-PCR.Results：The frequency of total CD4＋CD25＋T cells,CD4＋CD25highT cells and CD4＋CD25intT cells was not sinificantly different in those MRL/lpr Lupus Mice compared with that of healthy controls,But the index of CD4＋CD25highT：CD4＋CD25highT significantly and the expression of Foxp3 gene decreased in model mice,compared with that of healthy controls.The index and the expression of Foxp3 gene were significantly increased in the treatment groups compared with that of model group,especially the TCM Decoction plus west medicine group.Conclusion：The therapeutic mechansim of Compound Ziyin Qingre Prescription for SLE is probably related with the increase the level of protein expression of Foxp3,and regulate the balance of CD4＋CD25＋T cells subpopulations ratio,and induce the immunotherapy action to SLE.
