摘 要:目的 研究大鼠坐骨神经钳夹伤后碱性成纤维细胞生长因子(bFGF)mRNA表达的变化规律。方法 102 只Wistar 大鼠随机分为正常组6 只,对照及实验组各48 只,用持针钳钳夹坐骨神经,于伤后4 小时、1,3,7,10,14,21,28 天采用逆转录- 聚合酶链反应技术(RT- PCR) 结合聚丙烯酰胺凝胶电泳及alpha-32p- dCTP放射自显影,检测大鼠坐骨神经钳夹伤局部、腰4 ~6 背根节及相应脊髓节段bFGFmRNA的动态变化。结果 实验组伤后4 小时bFGFmRNA在损伤局部、脊髓及背根节的表达均开始增强,分别为1.41±0.10,1.78±0 .10 和2.17±0.12;伤后7 天表达值最高,分别为2.72±0.14,3 .65±0.17 和3.90 ±0.06;伤后28 天表达值为1.23 ±0.06 ,1.43±0 .05,1 .78±0.03,降至正常及对照组水平(P> 0.05)。Objective To study mRNA expression of basic fibroblast growth factor(bFGF) after crushing injury of sciatic nerve in rats. Methods One hundred and two Wistar rats were randomized into a normal group( n =6), a control group ( n =48) and a test group ( n =48). Sciatic nerve was crushed for 5 minutes using modified foceps. Reverse-polymerase chain reaction(RT-PCR) with polyacrylamide gel electrophoresis and alpha -32 P-dCTP labelling autoradiography were employed to detect the dynamic changes of bFGF mRNA in the lesion site, dorsal root ganglia of 4th-6th lumbar spine and spinal cord at 4 hours, 1, 3, 7, 10, 14, 21, 28 days after injury, respectively. Results bFGF mRNA expression of the test group began to increase in the lesion site, dorsal root ganglia and spinal cord at 4 hours (1.41±0.10, 1.78±0.10, 2.17±0.12), reached the peak at 7th day (2.72±0.14, 3.65±0.17, 3.90±0.06), returned to control levels at 28th day (1.23±0.06, 1.43 ±0.05, 1.78±0.03) after injury. Conclusions The increase of endogenous bFGF mRNA after peripheral nerve injury may enhance the regeneration of peripheral nerve.
