摘  要:采用植入后全胚胎培养方法研究重铬酸钾对大,小鼠胚胎的发育毒性. 结果表明,重铬酸钾在1.0 和2.5 m g·L- 1以上时分别可致大,小鼠胚胎体长,头长,卵黄囊直径及胚胎干重等各项指标明显低于对照组,同时观察到大,小鼠胚胎脑,神经管和体屈等发育迟缓,组织光镜可见胚胎神经管上皮变薄,细胞排列不规则. 以上结果均呈明显的剂量-反应关系,表明重铬酸钾体外对大,小鼠胚胎具有明显的发育毒性,且大鼠比小鼠敏感.Nine and a half day old rat embryos were explanted and cultured at six different concentrations of potassium dichromate in rat serum solutions (0, 0.25, 0.5, 1.0, 2.0, 4.0 mg·L -1 Cr) for 48 h. Rat embryo development in 0.25 mg·L -1 Cr was not significantly different from the control whereas the higher concentrations of Cr resulted in a concentration related decrease in yolk sac diameter, crown rump length, head length, embryo dry weight and Brown′s morphological developmental score. The 0.5 mg·L -1 Cr and higher concentrations also resulted in embryotoxicity as well as abnormalities, which included neural tube defect, deficient in axial rotation and fore limb, maxillary process defects. Eight and a half day old mouse embryos were explanted and cultured in 0, 0.6, 1.2, 2.5, 5.0, 10.0 mg·L -1 Cr rat serum for 48 h. Mouse embryonic development in 0.6 mg·L -1 Cr group was similar to that of the control. But all higher concentration groups had a significant decrease in development score, yolk sac diameter, head length, crown rump length and dry weight, the 1.2 mg·L -1 Cr and higher concentrations also resulted in embryotoxicity and abnormalities, which were similar to that of the rat embryo. These results indicate that K 2Cr 2O 7 is developmental toxic in both rats and mice but rats are more sensitive than mice.
