补肾方对自然衰老大鼠海马CA1区凋亡相关基因Bax、Bcl-2 mRNA表达的影响

摘　　要：目的:从自然衰老大鼠海马神经元凋亡角度探索补肾方的作用机制。方法:以自然衰老SD雄性大鼠(24月龄)为肾虚衰老模型,大鼠随机分老年组(模型)、左归组和右归组,另设青年组,共四组。大鼠自20月龄始,左归组和右归组分别给予左、右归丸,均按相当于生药7.5g/kg(成人临床用量的15倍)自由饮用稀释药液,每周停药2天,连续4个月;原位杂交检测海马区神经元Bax、Bcl-2 mRNA表达。结果:与青年组比较,老年组大鼠海马CA1区Bcl-2基因mRNA表达水平显著下降(P<0.05),Bax基因mRNA表达水平显著升高(P<0.05);与老年组比较,左归组和右归组大鼠海马CA1区Bax、Bcl-2基因mRNA表达水平均有显著变化(P<0.05)。结论:自然衰老大鼠的海马CA1区神经细胞凋亡相关基因表达异常,补肾方可通过调节海马CA1区神经细胞Bax、Bcl-2基因mRNA的表达,降低海马神经细胞凋亡,进而改善海马功能、延缓衰老。Objective：To study the mechanism of aging and Bushen Prescriptions through apoptosis of hippocamal neuron in natural aged rats.Methods：Natural aging Male SD rats were as shen abscence model（24 months old）.SD rats were randomly divided into three groups：old group、Zuogui group and Yougui group.Young group was setted independenly.Treatment with Zuogui Pill and Yougui Pill.From 20 months old the rats were given Yougui Pill and Zuogui Pill with 7.5g/kg for 4 months（two days with no pill every week）.Rito hybridization was used to detect the expression of Bcl-2 and Bax mRNA in CA1 of hipocampus in all groups.Results：Compared with young group,the expression of Bcl-2 mRNA were decreased and Bax mRNA were increased in old group（P〈0.05）;Compared with old group,the expression of Bcl-2 mRNA were increased and Bax mRNA were decreased in Yougui and Zuogui group（P〈0.05）.Conclusion：Expression of apoptosis related gene mRNA is disordered in hippocampal CA1 of natural aging rats.Bushen Prescriptions can ameliorate the state of apoptosis of hippocampus and function of hippocampus and antiaging through regulating expression of Bcl-2 and Bax mRNA in CA1 of hipocampus.
