加味四逆散对慢性应激大鼠额叶5-HT_2AR及其mRNA表达的影响

摘　　要：目的:探讨加味四逆散对慢性应激模型大鼠额叶5-HT2A受体及其mRNA表达的影响。方法:选择open-feild法评分相近的74只大鼠随机分为对照组、模型组、氟西汀组、加味四逆散大、中、小剂量组。应用分养和长期不可预见的中等强度应激造成慢性应激大鼠抑郁模型,以免疫组化法和原位杂交法研究大鼠额叶皮层5-HT2AR及其mRNA的变化。结果:加味四逆散各剂量组于治疗的第7d起效,但无统计学差异,于第14d水平活动、垂直活动和糖水消耗量均增加,且与模型组有统计差异,与对照组比,模型组额叶皮层5-HT2AR及其mRNA表达降低;与模型组比,各治疗组5-HT2AR表达均升高。除加味四逆散小剂量组外,各治疗组mRNA表达均升高。结论:慢性应激能诱发大鼠轻度抑郁障碍和额叶皮层5-HT2AR及其mRNA的低表达,加味四逆散改善慢性应激大鼠的抑郁症状可能与调节额叶皮层5-HT2A受体系统有关。Objective：To investigate the intervention of modified Sini Powder on 5-HT2AR and 5-HT2AR mRNA in frontal lobe of rats chronic mild stress model.Methods ：74 Sprague-Dawley rats were randomly divided into 6 groups.The models animals were given separation and chronic unpredictable mild stress.Immunohistochemistry and hybridization in situ were used to study the changes of 5-HT2AR and 5-HT2AR mRNA in the frontal lobe of rats.Results ：The 5-HT2AR and 5-HT2AR mRNA express were lower in model group than in control（P〈0.01）;The 5-HT2AR express were higher in treatment groups than model group（P〈0.05,P〈0.01） and 5-HT2AR mRNA express were higher except low dose modified modified sini powder（P〈0.05,P〈0.01）.：Conclusion The results indicate that the chronic unpredictable mild stress may induce the mild depression and decrease the 5-HT2AR and 5-HT2AR mRNA level in frontal lobe.The modified sini powder has the function to improve the depressive symptom in the depressive rats and its one mechanism maybe by effect the 5-HT2A receptor system.
